چکیده

Objective: MafA has been recognized as a specific pancreatic β-cells activator for Insulin expression. This study investigated the changes in glucose transport and consumption by MafA-positive Insulin-producing cells resulting from differentiation of rat adipose tissue-derived mesenchymal stem cells. Methods: Adipose were differentiated into 14-day protocol using nicotinamide ITS. The ability to produce was confirmed adipogenesis capacity expression Oct4, Nanog, Sox2. Glucagon concentrations determined ELISA. presence MafA, Glut2, ENO1, IGF-I receptor genes Western blotting analysis. amounts Glucokinase Pyruvate Carboxylase activities measured Fluorometric Colorimetric methods, respectively. Results: un-differentiated showed promising differentiate toward adipocytes. tissue revealed significantly elevated levels Sox2 compared that secreted considerably more than undifferentiated

In-vitro Differentiation of Human Umbilical Cord Wharton’s Jelly Mesenchymal Stem Cells to Insulin-Producing Cells

Background & Objective: Diabetes is a major chronic metabolic disease in the world. Islet transplantation is a way to treat diabetes. Unfortunately, this method is restricted due to graft rejection and lack of donor islets. Mesenchymal Stem Cells (MSCS) have the ability to differentiate into Insulin-Producing Cells (IPCs). In this study, Human Umbilical Mesenchymal Stem Cells (HUMSCS) were in...

effects of exendine-4 on the differentiation of insulin producing cells from rat adipose-derived mesenchymal stem cells

background: to evaluate the effect of exendine-4 (ex-4), a glucagon-like peptide 1 (glp-1) receptor agonist, on the differentiation of insulin-secreting cells (ipcs) from rat adipose-derived mesenchymal stem cells(admscs). materials and methods: in this experimental study, admscs were isolated from rat adipose tissue and exposed to induction media with or without ex-4. after induction, the exis...

Differentiation of Human Adipose Tissue-Derived Mesenchymal Stem Cells into Insulin Producing Cells Using Minimal Differentiation Factors

Background & Aims: Type 1 diabetes, or insulin-dependent diabetes, is an autoimmune disease in which pancreatic beta cells are destroyed by the immune system. Hitherto, no definite treatment has been found for this condition. Mesenchymal stem cells (MSCs) are multipotent, self-renewing cells that have the ability to differentiate into mesodermal tissues. This ability has attracted the attention...

Differentiation of Mesenchymal Stem Cells Derived From Human Adipose Tissue into Cholinergic-like Cells: In Vitro Study

Introduction: Cholinergic-associated diseases currently constitute a significant cause of neurological and neurodegenerative disabilities.  As the drugs are not efficient in improving the suffered tissues, stem cell treatment is considered an effective strategy for substituting the lost cells. Methods: In the current study, we set out to investigate the differentiation properties of human adip...

Differentiation Potential of Nestin (+) and Nestin (-) Cells Derived from Human Bone Marrow Mesenchymal Stem Cells into Functional Insulin Producing Cells

The feasibility of isolating and manipulating mesenchymal stem cells (MSCs) from human patients provides hope for curing numerous disease and disorders. Recent phenotypic analysis showed heterogeneity of MSCs. A nestin progenitor cell is a subpopulation within MSCs which plays a role in pancreas regeneration during embryogenesis. This study aimed to separate nestin (+) cells from human bone mar...
